Background :
The defective gene behind the INCL(infantile neuronal ceroid lipofuscinoses) disease, CLN1, encodes for palmitoyl protein thioesterase 1 (PPT1). It consists of 306 amino acids, including a signal sequence of 26 amino acids and three N-linked glycosylation sites. This protein exists with the molecular mass of 32 kDa, 34 kDa,36 kDa and 38 kDa. The enzyme is transported into lysosomes of non-neuronal cells by the mannose 6-phosphate receptor (M6PR) mediated pathway and a significant amount of native PPT1 resides in a complex rather than in a monomeric form. Cellular location Lysosome, Secreted

Antibodies are immunoglobulins secreted by effector lymphoid B cells into the bloodstream. Antibodies consist of two light peptide chains and two heavy peptide chains that are linked to each other by disulfide bonds to form a “Y” shaped structure. Both tips of the “Y” structure contain binding sites for a specific antigen. Antibodies are commonly used in medical research, pharmacological research, laboratory research, and health and epidemiological research. They play an important role in hot research areas such as targeted drug development, in vitro diagnostic assays, characterization of signaling pathways, detection of protein expression levels, and identification of candidate biomarkers.
