Background :
MAP2K3 encodes the mitogen-activated protein kinase 3 (MEK3) belonging to the mitogen-activated protein kinase (MAPK) signaling pathway. Dysregulation of the MAPK pathway is frequently observed in various cancers. MEK3 is a dual-specificity kinase that belongs to the MAPKK family responsible for the phosphorylation of serine/threonine and tyrosine residues of downstream MAPK substrates. Critical to the MAPK p38 signaling pathway, MEK3 and MEK6 activate p38 to induce cellular differentiation, migration, survival, apoptosis, and metabolism. Mice lacking MEK3 and MEK6 are more likely to develop cancer than their wild-type counterparts. Similarly, MEK3 expression is often downregulated in breast, colon, liver, esophageal squamous cell carcinoma, and thyroid cancer.
