E fHbp var1.1 appears to become the result of a cooperative and elaborate network of interactions. In spite of the sequence diversity inherent to fHbp, we show here that mAb 1A12 recognizes a series of fHbp variants with incredibly higher affinities, suggesting a higher breadth of coverage potentially conferred by this human mAb. Structure of no cost Fab 1A12 reveals paratope flexibility. We also determined the crystal structure of your free of charge Fab 1A12 at 1.76 resolution (Table 2). Comparison from the free of charge and antigen-bound Fab structures shows that they are very related (rmsd 0.69 on alpha carbons).
