Applications: ELISA:To detect hIL-3 by direct ELISA (using 100 μL/well antibody solution) a concentration of 0.25 – 1.0 μg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2 – 0.4 ng/well of recombinant hIL-3. Sandwich:To detect hIL-3 by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.25 – 1.0 μg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with our Polyclonal Anti-Human IL-3 (XP-5185) as a capture antibody, allows the detection of at least 0.2 – 0.4 ng/well of recombinant hIL-3.Western Blot:To detect hIL-3 by Western Blot analysis this antibody can be used at a concentration of 0.1 – 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-3 is 1.5 – 3.0 ng/lane, under either reducing or non-reducing conditions.

Background: The pleiotropic IL-3 (Interleukin 3) is a 15kDa cytokine that is primarily secreted by activated T lymphocytes and stimulates the proliferation and differentiation of hematopoietic cells. IL-3 acts in hematopoiesis by controlling the production, differentiation, and function of two related white cell populations of the blood, the granulocytes and the monocytes/macrophages. It induces granulocytes, macrophages, mast cells, stem cells, erythroid cells, eosinophils and megakaryocytes and is expressed by activated T cells, mast cells, and natural killer cells. IL-3 not only supports growth of both pluripotent stem cells and the more differentiated committed progenitors, but it also stimulates the functional activity of some fully differentiated cells. IL-3 has also been shown to protect mast cells from undergoing apoptosis.
