Background: BMP1 encodes a Metallopeptidase M12A type protease that is comprised of an amino terminal signal peptide, prodomain and an astacin-like protease domain followed by CUB and EGF-like motifs. BMP1 protein has been shown to cleave chordin, the C-terminal propeptides of procollagen I, II, III and VII, pro-lysyl oxidase, probiglycan, and laminin-5, and its expression has been shown to induce cartilage and bone formation in vivo. At least six spliced variants have been reported. These share the N-terminal protease domain but differ in the C-terminus, and thus contain varying numbers of CUB and EGF-like domains.
